Abstract:
Hepatocellular carcinoma (HCC) was one of the most common malignancies with high incidence and mortality which threatened human health. Integrating analytical technology of high throughput and high resolution with bioinformatics, metabonomics could investigate the organism on the level of metabolism, provide a unique insight into searching biomarkers for early diagnosis of HCC and exploring the mechanism of HCC. The advances of metabonomics studies on HCC in the recent years were reviewed in this paper which could provide the reference for further research.

2013, 31(2): 86-89,115. doi: 10.3969/j.issn.1006-0111.2013.02.002
Abstract(2953) PDF (575KB)(500)
Abstract:
Some unique inherent properties of polymeric micelles, including small particle size, high stability, long residence time, and good biocompatibility allowed polymeric micelles to be used as drug carriers. In recent years, increasing reports about the polymer micelles had been designed for tumor targeted drug delivery systems, including passive targeted drug delivery using tumor pathological nature and active targeting drug delivery using surface modification of polymer micelles. The research progress of polymeric micelles used as tumor targeted drug carriers were reviewed in this paper.

2013, 31(2): 90-93. doi: 10.3969/j.issn.1006-0111.2012.02.003
Abstract:
In biological samples, the concentration and efficacy of drugs had great correlation, especially in the plasma (or serum), the concentration of drugs was directly related to efficacy. There was a wealth of drug metabolites in the urine, which was also frequently used. Because saliva collection convenient and sometimes it had great relevance with the plasma free drug concentration, so it was used sometimes. Other organs and tissues, except the special needs, were less used. Because of the complexity and diversity of sample preparation techniques, drug analysis of biological samples put forward higher requirements. In a variety of treatment methods, solid phase extraction attracted more and more the attention from the majority scientists because of its applicability, practicality. The reports with the application of solid phase extraction (SPE) in the analysis of biological samples were reviewed, in order to provide theoretical guidance and technical support to the relevant researchers.

Abstract(4278) PDF (618KB)(334)
Abstract:
The aldehyde dehydrogenase II (ALDH2) could act as both esterase and dehydrogenase. The improvement of ALDH2 activity might reduce the damage caused by alcohol and ischemia and avoid the occurrence of some certain cancers. The outline of ALDH2 and its agonists were summarized concisely through literature analysis, collation and induction in this paper.

2013, 31(2): 98-101,107. doi: 10.3969/j.issn.1006-0111.2013.02.005
Abstract(2344) PDF (624KB)(584)
Abstract:
Objective To design and evaluate a multifunctional and multiple unit oral DDS, which depends on the preparations of pulsatile release mini-tablets. Methods Based on the combinations of pulsatile release tablets, zero-order DDS could be obtained. Velocity-time curve, instead of the cumulative percentage drug release profile, was plotted. The nonlinear least square model fit program was applied to process the velocity data of dissolution. The test curves coincided with the theoretical curves from simple summation of v-t equations of individual pulsatile release tablets. Results Zero-order DDS were designed with two kinds of combinations. The amount of the two pulsatile should be 1:1. The combination of pulsatile release tablet could achieved the formulation from which Acetaminophen and Pseudoephedrine could be released in the same proportion. Conclusion The combination of delayed-onset sustained-release Tablet was better fit in with zero-order equation and Acetaminophen and Pseudoephedrine could be released in the same proportion.

Abstract(2899) PDF (634KB)(136)
Abstract:
Objective To design and synthesize the new 2-pyridyl ketone amide derivatives and test the antiviral activity. Methods According to the pharmacophore model, a new class of 2-pyridyl ketone amide derivatives was designed, which were synthesized by Heck reaction, selective nitration reaction, catalytic hydrogenation reaction, acylation reaction and the open-loop rearrangement reaction. All the synthesized compounds were confirmed by 1H NMR, and their anti-HBV-DNA replication activity were determined. Results The designed novel 2-pyridyl ketone amide derivatives had inhibitory activity against HBV-DNA replication. Among them, compounds 6h、6e and 6a showed the best inhibitory activity. Conclusion When the 4-ethoxyl phenyl group was attached on the N atom of the benzyl ring, the compounds revealed good inhibitory activity.

Abstract:
Objective To established a two-step discriminant method based on SVM and HQI or PCA, in which cephalexin capsules from several domestic manufacturers were used as tool drugs for counterfeits of brand drugs appear on the market. Method In the first step, a simulating classification model of cephalexin brand capsules and 4 kinds of counterfeit drugs was built, the cross-validation accuracy was 95.63%, and prediction accuracy of suspicious samples within the model was 93.75%, and the false-positive rate of additional homemade test samples was 25%. In the second step, the positive samples were applied to HQI and PCA methods, and the false positive rate could be reduced to 5% and 0% respectively. Results and Conclusion The two-step discrimination method established in our study could detected counterfeits of brand drugs efficiently and accurately.

Objective To evaluate the efficacy and explore the mechanism of Jiangtang Mixture in treatment of diabetic peripheral neuropathy (DPN). Methods 60 cases of DPN patients were randomly divided into treatment group and control group with 30 cases in each group. Two groups were all treated with a diet control, routine hypoglycemic therapy, the control group was treated with Methylcobalamin Tablet 0.5 mg, po, tid. While the treatment group was treated with Jiangtang Mixture 50 ml, po, tid, on the basis of the control group. The course of treatment was 4 weeks in each group. Before and after treatment, clinical signs and symptoms were observed and determined, including fasting plasma glucose (FPG), the 2 hours postprandial blood glucose (PPG2h), glycated hemoglobin (HbA1C), plasma viscosity (PV), endothelin (ET), nitric oxide (NO), and nerve conduction velocity (NCV). Results The total effective rate of the treatment group (86.67%) was significantly higher than that of the control group (63.33%)(P<0.05). There were significant differences in levels of the indexes, such as NCV、FPG、PPG2h、HbA1C、PV、ET and NO in the treatment group before and after treatment (P<0.05), but only in the four indexes, such as NCV、PV、ET and NO, the treatment group compared with the control group showed significantly differences after treatment (P<0.05). Adverse reactions of Jiangtang Mixture did not find in the observation period. Conclusion Jiangtang Mixture combined with Mecobalamine to treat DPN was safe and effective. The mechanism could be related to protect vascular endothelial cell, improve blood rheology, expand the peripheral microvascular, nourish the nerve with blood and oxygen supply, prevent or reverse impaired peripheral nerve fibers of DPN.
